Changing the kinjflow will need the VE table scaled. Changing it from 0.0566 to 0.045 then to get the tune roughly back to where it is now multiply every value in the VE table by 1.25 (you can do this in TP using the math functions).

But the VE is going to change at low loads once enabling single fire, and its currently crazy rich above idle which may or may not be the misfire issue, so I think yes you will need to get it tuned again. Its a long way off still, to start with just go through getting the target to match the wideband in areas you can get to in neutral and holding it at various RPMs.
