Thats a very generic question - What interface module do you intent to use? If you just want to use a 6 cyl reluctor distributor, its really just cylinders set to 6 and the low res crank flag in the platform options, but you'd be better off starting with a 6 cylinder base OS that is set up like this. You will loose sequential and loose lots of your ignition options but it'll run it.

Its also been shown that you can use these settings and interface a commodore ignition module to retain coilpacks, but nobody who has done it has confirmed exactly how back to the forums. You can see where this conversation got to last time here: viewtopic.php?f=42&t=5023

Hi all:

People need to remember that the 0411 came on the 4.3l V6 from 2001 onwards in the GM S-series trucks and Full sized trucks (yes it was superseded by the P59 later on).
Would be nice to be able to upgrade a FWD V6 with one!

Hi Antus, I have been involved in conversion and this was done. I have some notes (somewhere) on how it finished up and a copy of the calibration.

Hi mate, just what we need someone hands on doing it.
Maybe get me on messenger and we can work through it and make sure I'm right. I didn't tune it but had confirmation it ran.
I have a copy of the calibration after the dyno tune (somewhere)
